Intrathecal (IT) drug administration bypasses the blood-brain barrier by injecting therapeutic molecules directly into the central nervous system. Infusion experiments demonstrate the intrathecal route as a viable method for targeting molecules to the spine or the brain. Multimodal imaging confirms that IT administration with proper infusion settings can achieve wider dispersion than has been reported previously. Validation by in vivo nonhuman primate data confirms our ability to predict the biodistribution of intrathecally administered agents from first principles.
